Preparation and Characterization of Calcein Entrapped Ethosome

Abstract

Calcein entrapped ethosomes, which are vesicular system composed mainly of phospholipid, ethanol, and water, were prepared with varying the composition of constituent comonents. Liquid crystalline phase behavior and thermal phase transition of the ethosomes were investigated by the polarized optical microscopy equipped with hot stage. Average droplet sizes of the ethosomes, ranged from 50 nm to 1 μm, depended on lipid composition and sonication time. Compared with liposome, ethosomes showed much higher entrapment efficiencies, which were dependent of lipid composition, amount of added calcein solution, and preparation method. Stability changes of ethosome membrane with temperature, lipid composition, and storage time were verified through the calcein release test. Calcein release was observed to be less than 5% at 45 ℃ for 30 days, while most of calcein was released at 60 ℃. Incorporation of cholesterol into ethosome delayed calcein release at high temperature.

초록

인지질, 에탄올, 물로 구성된 소포체인 ethosome을 구성성분을 달리하고 수용성 지표물질로 calcein을 포집하여 제조하였다. 온도조절장치가 갖춰진 편광현미경을 사용하여 ethosome의 액정상 거동과 열적 상전이를 관측하였다. Ethosome 입자의 평균 크기는 지질조성과 제조과정 중 초음파 처리시간에 따라 50 nm에서 1 μm까지 다양하게 나타났다. 리포좀과 비교할 때 ethosome은 훨씬 높은 포집효율을 나타냈고 지질조성, 첨가되는 calcein 수용액양, 제조방법 등에 따라 포집효율이 달라진다. 온도, 지질조성, 저장시간에 따른 ethosome 막 안정성의 변화를 calcein 방출 실험을 통해 확인하였다. 45 ℃에서 30일 동안 calcein 방출률은 5% 미만이지만, 60 ℃에서는 calcein이 거의 다 방출되었다. Ethosome에 콜레스테롤을 첨가하면 고온에서 calcein 방출이 지연되었다.
